Will the B5 S4 rotors work on a B5 A4 for the front and rear?

Not without have the pads and calipers too. You may also need an adaptor for the front clipers.

is there a greater advantage switching over to the S4 brake, caliper, and rotors upgrade?

bigger brakes = better stopping power.

You may also need an adaptor for the front clipers.
You do need the front caliper carriers.
