Yes they can. I assume you mean a tiptronic transmission.
I had the APR chip 93 octane and a 710N diverter valve installed last week. It is like driving a whole new car. I've gotten my moneys worth just in the last 7 days.

There is different programming for the tiptronic versions, but yes they are just as effective as the software for the manual.
